Intended use

This ELISA kit applies to the in vitro quantitative determination of Human α2-PI concentrations in serum, plasma and other biological fluids.

Detection Range

12.5 to 800ng/mL.

Sensitivity

The minimum detectable dose of Human α2-PI is 7.5ng/mL

Specificity

This kit recognizes natural and recombinant Human α2-PI.

No significant cross reactivity or interference between Human α2-PI and analogues was observed.

Repeatability

Coefficient of variation were<10%.

Test principle

This ELISA kit uses SandwichELISA as the method.

The micro ELISA plate provided in this kit has been precoated with an antibody specific to α2-PI.

Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody.

Then a biotinylated detection antibody specific for α2-PI and AvidinHorseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated.

Free components are washed away.

The substrate solution is added to each well.

Only those wells that contain α2-PI, biotinylated detection antibody and AvidinHRP conjugate will appear blue in color.

The enzymesubstrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow.

The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm.

The OD value is proportional to the concentration of α2-PI.

You can calculate the concentration of α2-PI in the samples by comparing the O.D. of the samples to the standard curve.
